In certain instances, insertion of a DNA fragment into such cleavage sites blocks expression of kanamycin and/or tetracycline resistance while insertion of other fragments permits expression to occur.
The reasons for this are not clear; it may be simply that the extrachromosomal state of plasmids provides more opportunity for genome-genome interaction, but we don't know that.
